If you took RNA polymerase out of the equation? Human cells use RNA polymerase to make a template of a cell’s DNA. It is one of the first steps in the process of eventual protein production. RNA is the enzyme that makes RNA chains using genes found in DNA as a pattern. These specific genetic codes are  expressed on mRNA. When the toxin blocks the action of RNA  polymerase, it by default, stops the production of mRNA in the transcription phase. mRNA, if available, goes on to the translation phase and is read by tRNA. In the reading of the mRNA pattern, tRNA matches the correct amino acid to that specific pattern. The amino acids are read and placed three at a time to match the mRNA codon. From these codon of amino acids, polypeptides are formed with the help of the cell’s ribosome. In turn, these polypeptides form to make proteins. (Hudon-Miller, 2012) What’s the big deal about protein? Proteins, the performers of the cell, cause action at all levels of cellular functioning. At the cellular level protein is needed to reproduce that cell, for cell structure and functioning. Proteins are also used as enzymes to begin other biochemical functions that affect everything from immunity to electrolyte balance. The liver’s function has decreased because the proteins needed to make it’s cells work have been blocked by alpha-amanitin’s ability to stop protein production simply by blocking the function of RNA polymerase. These proteins are so important that without them you die as they destroy first your body’s filters, the liver and the kidneys. Mohawk Indians: Past and Present Essay

The Mohawk Nation is a Native American tribe of the New York area. They were a sedentary tribe who practiced agriculture in the harsh northeast climate. The primary crops were corn, squash, and beans. The Mohawk were skilled trappers who took advantage of this skill when the Europeans arrived in their area. The tribes worked with other tribes to achieve better relations with other Native Americans and Europeans. This included a constitution and treaties with Americans and Canadians. The modern day Mohawk Nation has tried to keep their culture and their land. The nation of the Mohawks once covered a large area of New York, Ontario, and Quebec. Present day Mohawks mainly live on three reservations. The tribes on these reservations are the Akwesasne, Ganienke, Kahnawake, Kanesatake, Kanatsiohareke, Six Nations, Tyendinega, and Wahta (Mohawk Nation Council of Chiefs, 2007). The tribe deals with many modern day problems that are similar in nature to the issues of the United States as a whole. Education is a concern for the tribe. The main issue of education is the focus on retaining their culture and obtaining the level of education needed in today’s society. Their answer to this problem was to found the Akwesasne Freedom School. This school was founded in 1979 by parents of the tribe for children in pre-k through eighth grade (Mohawk Nation Council of Chiefs, 2007). Besides the basic studies the curriculum focuses on culture beliefs and also includes a Mohawk language immersion program. Another concern of the Mohawk nation is the disputes of land ownership in their territories. The tribes of many areas in the New York and Canada area are fighting to keep rights to land that were allotted to them centuries ago. According to Connie Kidd, (2007) after the American War of Independence, during which Mohawk warriors fought as allies of the British, the Haudenosaunee were persecuted in their New York territories and so moved north to ancestral territories along the Grand River, which were reserved for them in the Haldimand Proclamation of 1784. This land was gradually taken out of their hands through disputes and surrenders. The tribes have had trouble being successful in getting the two governments along their  borders to honor separate treaties regarding land ownership. As in any other nation, crime is a concern for the Mohawk Indians. In the Mohawk territory smuggling has become a concern for the local citizens. Drugs, other contraband, and illegal border crossing have risen in the recent years. The smuggling problem seemed to have risen with increased taxes on tobacco in Canada. The smugglers, who are Mohawk Indians and other races, used the Akwesasne area to transport tobacco to Canada, so much so that the area became known as smugglers alley (Bonaparte, 2002). Another factor in the rise in smuggling is the duties that must be paid to bring goods into the Canadian side of the border. Since some of the Mohawk territory is divided between the Canadian and the United States borders the residents must pay to bring goods from one side to another. This has proven to be a hardship for the Mohawk Indians, especially in conducting business inside the reservation. The case of Mitchell vs. the Minister of National Revenue was a decision that stated the Mohawks of Akwesasne did not have an aboriginal right to bring trade goods across the border without paying duty (Bonaparte, 2002). This Canadian court decision, which itself took many years to decide, has affected the Mohawk Indians in conducting business inside and outside the reservation, and thus led to an increase in smuggling. The Mohawk Indians have had trouble combating this problem since the 15-member Tribal police force currently pays for border enforcement from its own operating budget that is supported by Tribal revenue (Indian Times, 2006). The tribes answer to help solve their crime problems is to train local Mohawks to serve in law enforcement. According to the Indian Times newspaper these tribal police officers are ideally suited to monitor this stretch of border as they are Akwesasne residents with an intimate knowledge of the territory (2006). The Office of Law Enforcement Services is responsible for improving law enforcement services and preserving the public’s safety throughout Indian Country. The United States has dealt with smuggling across the borders for many years and from every border. Prohibition times seen the smuggling trade rise in  this very same area yet the items were being smuggled into the United States instead of out. The methods of controlling this problem have changed since that time in history. Mohawk elders can recount that border patrol agents used deadly force to combat the smugglers, native and non-native alike (Indian Times 2006). Even though the smuggling issue has been present since the prohibition days in the early twentieth century the United States government is only now beginning to understand the impact it is having on the Mohawk and surrounding communities. In 2006 the Bureau of Indian Affairs awarded the Tribal Police Department with a grant to help combat the drug smuggling and the illegal border crossing problem in Akwesasne (Indian Times, 2006). This funding is intended to increase the number of native officers to help fight the crime in the community. The Department of Justice and the Department of Homeland Security are among other government offices that have awarded grants to the police department to aid in buying equipment and addressing other issues (Indian Times, 2006). By working together the two nations can fight an ever growing problem and keep the Mohawk and surrounding communities safe. The various funding from the United States government can be effective in financing the programs needed to protect the community. Aristotle and Plato’s Concept of the Soul Essay Most individuals are interested in knowing and understanding the various phenomena that exist among their midst. It could be as simple as looking for information as to why the sun rises and sets or it could involve very trivial matters that deal with things that is not seen by the naked eye. This is why the field of philosophy exists in order to investigate the ideas of truth, existence, knowledge, and other subjects by means of logic and reason (Kemerling). In relation to this, many famous philosophers have their respective theories that tend to explain various matters. This could be exemplified by Aristotle and Plato’s ideas behind the existence of a soul. Aristotle wrote three books of De Anima wherein he elaborated a varying array of philosophical and scientific topics. In Book II, Chapter 1 of De Anima, Aristotle made a sketch or outline of the nature of the soul. He explained the concept of the soul by using a scientific perspective that uses elements of biology. In relation to this, he also employed the idea of metaphysics that tackled everything such as substance, form, matter, potentiality, and actuality. Aristotle perceived that the soul is united with the living body. As such, the existence of the soul is also dependent upon the host. He deems that the soul is made simply for the purpose of development, which can only happen if it is connected with a body or some kind of container in the physical world. In this sense, the soul is assumed to exist as the form of the body. The importance of the soul is greatly dependent upon the body or a type of entity that gives life to it. Since the soul designates life, Aristotle believed that it is also present in every living thing, including plants and animals. He also elaborated that different entities have various versions of the soul. The soul of a human being is unique from others because it has the ability to hold rational beliefs and use reason. It is regarded as the higher level of soul while the lowest ones are those that are found in plants and animals. Moreover, Aristotle’s association of the soul to specific forms made the soul as the mover because it is the culmination of the various life forces (Davidson). On the other hand, Plato believes that the soul is made up of three basic energies, including reason, emotion, and appetite that are responsible in animating human beings. The energy of reason is regarded as having the greatest value, while emotion and most especially appetite are considered as the lower passions. Reason is responsible in governing the soul that controls the emotion and the appetite of an individual. In relation to this, Plato deems that the soul is important in living a moral life. He explained that morality is the cause of happiness, which motivates an immoral person to behave righteously if he or she wants to be happy. Plato asserted that a happy person is a just person. Moreover, the psychic harmony of the soul is expressed in four cardinal virtues that are related to the three basic energies. In terms of reason, the just person has wisdom or prudence. In relation to emotion, the just person has the virtue of courage. Lastly, when it comes to the energy of appetite, a just person has the value of temperance (â€Å"Plato theory of the Soul†). Aristotle and Plato’s respective theories of the soul have similarities in the sense that they both believe that the soul is responsible for the ability of human beings to think logically and rationally. Nevertheless, there are many differences in their concept of the soul. Since Aristotle employed the scientific topics in his theory, he perceived the soul as dependent upon the body. In relation to this, he also deemed that other living things have souls. On the contrary, Plato merely focused on the existence of human beings’ soul. He explained his idea of the soul in the light of morality rather than biology. Furthermore, Plato argued that the soul is influenced by external force in order to move, while Aristotle asserted that the soul is the mover itself because it is an assortment of life forces. The varying theories of Aristotle and Plato with regards to the soul only show that philosophers have different perspective and understanding in the pursuit of understanding various matters of an individual’s existence.

Pleasantville Essay The movie â€Å"Pleasantville† (Produced and directed by Gary Ross in 1988) is more than just your ordinary film. Pleasantville sends a message within a message. Not only is it a great film, (made popular by New Line Cinema) it contains numerous biblical references. Some of the biblical references in this film may be easier to see and understand than others. But when asked to analyze this film and critique the biblical references you will understand just how many there are. To me the most obvious relation to the bible is in a scene where the main character David (Tobey Maguire) is the first to see a fire that breaks out in a tree in his front yard. Ironically enough, the tree is on fire, but is not consumed by the flames, it’s burning but not disintegrating. David then ran to the fire and distinguished it while commanding the fire fighters on what to do. How does that relate to the bible? In Exodus 3: 1-15 is the story of the burning bush. The burning bush was also on fire and not consumed by the flames, this was when God appointed Moses to lead the Israelites out of Egypt. Much like how David was then appointed a leader of Pleasantville. The only real difference is that the biblical story says that the burning bush talks to David. (The burning bush being God). Another obvious biblical relation in the film goes along with the story of Adam and Eve. The biblical version of the Adam and Eve (Genesis 2 and 3). story says that God created man (Adam) and placed him in the Garden of Eden to work and watch over it. God allows Adam to eat from all trees except from the â€Å"tree of knowledge of good and evil†. God tells Adam â€Å" for on the day you eat from it, you will surely die†. Later on in Genesis 2, Adam needs a help mate to look over the garden, but none of the animals in the garden are to Adam’s satisfaction, so God then makes a woman (Eve) out of Adam’s rib while he is asleep. On a side note, Adam and Eve are both naked and neither one of them are ashamed of themselves. Finally the serpent is introduced in Genesis 3, the serpent is said to be â€Å" slier then every beast on the fieldâ€Å". The serpent eventually persuades Eve to eat from the â€Å"Tree of Knowledge†. After she eats from it, she has Adam eat from it. Shortly after, they finally notice they are naked and become self conscious and cover up. God then asks about what they have done and Eve blames Adam, while Adam blames the Serpent. God curses them, and makes Eve suffer through labor and banishes both of them from the Garden of Eden. The relation in the movies comes from a scene in the end involving David and another girl from his school. Color starts to fill Pleasantville, and the girl he is sitting with in the lawn gets up and grabs a fruit of off the tree, and then she hands the fruit to David to eat, much like Eve did with Adam. As soon as they ate the fruit, it started to rain in Pleasantville for the first time ever, symbolizing God’s anger, and color now completely filled Pleasantville. The discovery in books also reminds me of the â€Å"Tree of Knowledge of good and evil†, the books being good, and the reading of them being evil because it made people curious to as what else was out in the real world. Therefore there is an effect in both cases, the biblical one, and also the movie. Pleasantville, (before David and Jennifer) is a â€Å"dream world† if you will, everyone is always happy and there are never any conflicts with anyone. Once the two of them arrive however, everything changes. In Pleasantville everything is black and white, but after Jennifer has sex with someone in Pleasantville, the first sighting of color comes out. To me, this symbolizes sin. The more people sin in the movie, the more color there is. So this biblical reference doesn’t necessarily correspond with any specific part of the bible, just sin in general. The people that sinned were turned into color, and at first if you were colored, you were frowned upon. But as more and more people â€Å"sinned† it became the â€Å" new norm†. So at first sinners were looked at in a negative way, but as more and more people sinned it became okay, and there was a neutral outlook on them. If I had to match a few characters in the movie to people in the bible I would say that Betty Parker (housewife) is most life Eve. I say this because at first she is so innocent and has no idea what sinning (or having sex) is. Then when she finally did sin she didn’t even understand or realize what she was doing was wrong. Jennifer ( Mary-Sue) to me, is most closely related with Satan. I say this because she was the one that first had sex (sinned), and she also spreads the knowledge of evil (sex). For example, in the movie Jennifer teaches her mother how to please herself, and she was also the first one to bring sex to Pleasantville. I would also say that David reminds me most of a Prophet because he is sent to Pleasantville and all in all he â€Å"delivers a message† to say the least. Another biblical allusion in the film that most people do not catch, would be the scene where David, and the soda shop owner paint a mural on the wall. They are expressing themselves, but, after doing research and seeing a picture of the mural that they made, I noticed something. On the left side of the mural there is a picture of a snake wrapped around an apple that is half eaten. This reminds me of the Garden of Eden and the serpent persuading Adam and Eve to eat the apple. At first glance you may not notice this, but after looking at the picture you can. The snake comes off us a very sneaky creature, much like the Serpent and how he tricked Even into eating from the Tree of Knowledge of Good and Evil. I don’t think that the film maker (Gary Ross) necessarily had an agenda when making this film, I think he just put in all these biblical allusions in the film to make people think. If I were to watch this film without trying to draw a connection with the bible, I wouldn’t have noticed any reference to it at all.

Sri Lanka is an island country and god has given to Sri Lanka blessings of nature blue seas, green hills, sandy beaches, abundant wildlife, cascading waterfalls, a riot of flowers and fruits, coconut groves, tea estates, spice gardens. Here ones day can be gloriously lazy by a palm cool lagoon or full of joy, swimming, snorkeling, and fishing or sailing like a brilliant jewel in the sparkling waters of the Indian Ocean. Sri Lanka is located in a strategically considered naval spot on the world map. This significant situation creates a path for a major sea route. In addition it creates a strategic naval link between West Asia, Africa and East Asia. Sri Lanka entered the international tourism arena in the 1960s. Since then, government involvement has been the key factor in tourism development in Sri Lanka. The Ceylon Tourist Board (CTB) was established in 1966 in order to provide direction and leadership to this promising sector of the developing economy. The country is famous for h ealthy and cultural heritage. Hospitality, tropical forests, natural and fascinating beaches, archeological sites and the tropical climate are the more attractive features. These factors make Sri Lanka a wonderful tourist destination. Over the years tourism in Sri Lanka has developed significantly. Today tourism has become the sixth Foreign Exchange Earner (FEE) in Sri Lankan economy. 2. Tourism can make a great impact on development of the Sri Lankan economy. Tourism is a fast growing industry which has been identified presently. Employment opportunities and regional development are playing significant role with regard to the tourism industry development. It can be considered as the most important industry in the country as a whole. The governments vision is to make the tourism sector as Sri Lankas most innovative and profitable income generating source. 3. During past three decades Sri Lankas tourism industry had few draw backs. This was mainly due to the security situation which prevailed in the country. Further the tourism industry was also affected due to the Tsunami catastrophe which occurred in 2004. Approximately 2/3rd of the coastal area including tourist infrastructure facilities was destroyed. The immediate post war period gave rise to a dramatic increase of tourist arrival from end May 2009. 4. In order to support the efforts and to develop the tourism sector in Sri Lanka there should be hotels with exclusive facilities which are in par with the international standards to accommodate a large number of visitors. In the year 2010 approximately one million tourists visited Sri Lanka. Unlike in the past, the inflow of tourists is increasing day by day as a result of the end of the 30 year civil war which destroys the whole country. Following graph indicates how tourism increased from 1966 to 2011 in Sri Lanka. The government is expected to receive 2.5 million tourists in 2016. To cater for this increase tourist hotels in Sri Lanka must be developed. Similarly other facilities should also be increased to meet the desired economic goals. AIM 5. The aim of this paper is to analyze the development of Sri Lanka tourism -challenges and opportunities GEOGRAPHY AND CLIMATE OF SRI LANKA 6. Sir Lanka has a various terrain but it mainly consists of flat lands but south-central portion of the countrys step sided river, canyons interior features and mountain. The compliment regions are the areas where most of Sri Lankas agriculture takes place, aside from coconut farms along the coast. Sri Lankas climate is tropical and the southwestern side of the island is the wet test. . The northeastern side of Sri Lanka is drier and most of its rain falls from December to February. Most of the rain in the southwest falls from April to June and October to November Sri Lankas average yearly temperature is 86 °F to 91 °F. An important geographic note about Sri Lanka is its position in the Indian Ocean, which made it vulnerable to one of the worlds largest natural disasters. On December, 26, 2004, it was struck by large tsunami that hit 12 Asian countries. Around 38,000 people in Sri Lanka were killed during this incident and much of Sri Lankas coast was destroyed. 7. Sri Lanka is situated in strategic location near major Indian Ocean sea lanes. It has a total area of 65,610 km ², with 64,740 km ² of land and 870 km ² of water. Its coastline is 1,340 km long. Sri Lankas climate includes tropical monsoons: the southwest monsoon (June to October) and the northeast monsoon (December to March), its terrain is mostly low, flat to undulating plain, with mountains in the south-central interior. The highest point is Pidurutalagala at 2,524.13 m. Resource include limestone, mineral sands, gems, graphite, phosphates, clay, and hydropower. 8. Adams Bridge, a land connection to the Indian mainland, is now mostly underwater with only a chain of limestone shoals remaining above sea level. That figure is projected to increase to 21,000 rooms by 2004, which will accommodate the target of 1 million tourists. Five regional domestic airports to be restructured at a total cost of approx. Rs. 2bn: Rathmalana, Koggala ,Ampara , Trincomalee Jaffna (Rathmalana will be developed as a City Airport) Previous year. This denotes that tourism industry is one of the core sources of foreign exchange earner in economy of Sri Lanka. Common political factors which affect the tourism industry are revolution, civil war, factional conflict, ethnic violence, religious turmoil, widespread riots, terrorism, nationwide strikes, protests, cross- national guerrilla warfare, world public opinion, repatriation restrictions, bureaucratic politics, leadership struggle, high inflation, border conflicts, high external debt service ratio and creeping nationalization. Last thirty years Sri Lanka also faced the civil war and it affected the tourism industry of Sri Lanka. Organizing elections frequently also create threat to the tourism industry because during the election period in some places to control the situation cur hews are implemented. 21. This will create negative image in the mind of tourists. Some of the foreign countries have reject Sri Lankas products in their country due to the violation of human rights during the war time. For example government invested and spent on IIFA programme but it was not a successful event for Sri Lanka because the big stars have boycotted the event. They didnt visit Sri Lanka because of the protests held in their country by asking them not to visit the place. Currently Sri Lanka is having negative relationship with some foreign countries therefore the export and import of Sri Lankan products will be decreased and it affects the tourism industry as well. FUTURE TRENDS, CHALLENGES 22. Source markets in Asia and Middle East to drive demand: Sri Lankas proximity to source markets such as India and its connectivity to the Middle East and China will help in sustaining tourism growth. The rise in per capita income and therefore consumer spending in these source markets will aid the growth in tourism. 23. Infrastructure growth to fuel tourism growth: To ensure rapid growth of tourism in the future, tourism projects will need to be balance with infrastructure development. We expect that with improvements in road infrastructure and development of the new airport in Hambantota, various new destinations will emerge in the country, especially to the north and the east, as these areas have abundant natural beauty to attract tourists but currently suffer from poor connectivity. We expect more airlines to operate in the country as the second airport develops. 24. Greater competition will facilitate growth: We also expect greater competition with the introduction of new hotels in popular destinations such as Colombo, Kandy, Bentota, and Sigiriya, and Galle. We expect the existing hotels to undertake phased renovations to effectively compete with the upcoming hotels. Also, with the increase in income from hotels over the past few years, we expect an increase in the number of hospitality related transactions and mergers and acquisitions. 25. Challenge of attrition to other destinations: Currently, the hotel industry witnesss significant employee attrition to countries in the Middle East and to the Maldives and India. However, with a more stable political environment and improvement in quality of living in Sri Lanka, we expect the attrition to gradually 26. Shortage of skilled labor: As new players enter the market, the projected growth in tourism will be accompanied by shortage of skilled labour. We anticipate the need for relive hospitality education institutions as the contribution of the tourism sector to the overall economy increases. SUMMARY 27. To ensure rapid growth of tourism in the future, tourism projects will need to be balance with infrastructure development. We expect the existing hotels to undertake phased renovations to effectively compete with the upcoming hotels. We also expect the hotel companies, especially the domestic companies located in the country to undertake long and medium term hospitality training programs and regularly undertake competition benchmarking for compensation to retain the talent. RECCOMONDATIONS 33. These are the recommendations which we can produce in related to tourism. Develop a marketing strategy: Emphasizing the creation and sustaining of a positive image of Sri Lanka as a substantive and well-established tourist destination offering facilities experiences that are distinct Sri Lankan products. Establish a domestic tourism strategy: To expand the existing product base and encourage a greater level of holiday movements in addition to pilgrim movements. Develop goods and services: To meet the increasing needs of the tourism industry, and thus optimize the economic benefit of tourism development to Sri Lanka. Modify the organization of tourism activities: In order to bring about related improvements in the private sector, increase coordination between tourism and interrelated sectors, and introduce streamlining to facilitate the development of the tourism sector. Emphasize the environmental, social and cultural aspects: Tourism Development Planning is the only way to solve many issues related to tourism in development. Therefore the tourism planning process should be designed to produce goals and objectives for the destination area related to tourism development. The tourism development plan provides overall guidelines for development and identifies development opportunities. Many development countries are involved in tourism planning. Financial Assistance: Financial assistance for hotel owners and any other profession in tourist sector would help to create more employment opportunities. Domestic Airport: Airport also more than 100 km away from the tourist destination it is needed have domestic airports. Surfing Training: This will create more employment opportunities and generate good income. Among the tourists there are some people they love for surfing and keen to learn. Conduct Language Teaching: Language barrier should be overcome by conducting specially job oriented language training. Employ Combat Divers: Surfing is risky and adventure sport it dont take much time for some to happen therefore as precautionary measure it is needed some combat divers close to the surfing area it would enhance the confidence of people who do surfing. Proper Taxi Service: Taxi service should be reformed introducing good vehicles fixed with meter system and giving training for drivers. Improve Infrastructure Facilities: Infrastructure facilities such as Power, Highways, Railways, Airports, Ports, Water Supply, Telecommunications, Accommodations, and Recreational, Shopping and Banking to be improved. Ethics - Virtue of Patience Essay -- essays research papers

Virtue and Patience In order to survive these days, there need to be at least some standards and goals of morality in a person's life. Moral excellence is definitely hard to achieve, but is something worth while to attempt. Personally, there are many distractions and obstructions the present day provides, creating a difficulty of direction in my own life. I need to overcome obstacles such as peer pressure in order to become a confident, successful person. The present day provides impediments which I believe I can overcome by way of morality and virtue. I have chosen to work on more patience in my life. According to Aristotle, for every virtue there are two vices. One is in excess and the other being deficit. Patience is a deficiency in my life. In having barely any patience, I am often stressed out and make more of simple situations. Aristotle would call this vice impatience. On the other end of the spectrum, the excess of patience is being passive. Aristotle and Plato, it was all about having balance in your life. I believe this theory is still in tact today. What I am striving for is not passivity, it is a moderate amount of patience, not too much and not too little, but just enough to make me more of a moral individual. The hard part about being virtuous is not practicing virtue sometimes but mastering it and doing it consistently. I have always been impatient and it would be extremely difficult for me to master being patient. One of Aristotle’s points about being a virtuous person is not doing everything well but doing what you do well and doing it consistently. Patience is defined as â€Å"good natured tolerance of delay or incompetence.† (www.dictionary.com) for me, this definition does the word a world of just... ...s outside and we ended up waiting for and hour for him to come back. I was going to flip out and tell my mom I wasn’t going to do this and that we should just leave and come back tomorrow but I remembered this essay and my goal of becoming more patient. So I put on my headphones and I went for a walk to find my cat, when I finally found him I was so excited for myself, I really felt like I accomplished something. For me this was a gratifying process and I actually enjoyed the journaling and paper. I think I could have put a little more effort into the specific instances. But over all it was alright for me. Before this I would have never noticed what lack of patience I had and how annoying it must be for the people around me. I’m excited to continue being a more virtuous person and improving my character through practicing more patience.

However, it brings challenges to HR specialist in JPNT as it is not a culture of sharing.
